Kirby Neurobiology Center, Boston Children&#8217;s Hospital <\/strong><\/div>\n<\/div>\n<p>Elizabeth Engle is Director of the F.M. Kirby Center and Program in Neurobiology at Boston Children&#8217;s Hospital, Professor of Neurology and Ophthalmology at Harvard Medical School, Investigator of the Howard Hughes Medical Institute, and Associate Member of the Broad Institute of MIT and Harvard. At Boston Children&#8217;s Hospital, she is a member of the Departments of Neurology, Ophthalmology and Medicine (Genetics), a member of the F.M. Kirby Neurobiology Center and the Program in Genomics, and a senior investigator for The Manton Center for Orphan Disease Research. Dr. Engle received her B.A from Middlebury College and her M.D. from Johns Hopkins University School of Medicine. She trained in pediatrics at Johns Hopkins, in neuropathology at Massachusetts General Hospital, and in adult and child neurology in the Longwood Neurology Training Program and at Boston Children&#8217;s Hospital. Following her residencies, she was a research fellow with Louis Kunkel, Ph.D., and later Alan Beggs, Ph.D., in the Division of Genetics at Children&#8217;s prior to establishing her own research lab in 1997. Her work has defined the human congenital cranial dysinnervation disorders and has been recognized by high-profile publications and by receipt of multiple honors, including the E. Mead Johnson Award for Research in Pediatrics from the Society for Pediatric Research, Sidney Carter Award in Child Neurology from the American Academy of Neurology, Research Award for Vision from the Alcon Institute, and the Bernard Sachs Award from the Child Neurology Society and election to the National Academy of Medicine and American Academy of Arts and Sciences. In addition to her research, Dr. Engle continues to care for patients, primarily consulting for children and adults with rare eye and facial movement disorders and other cranial nerve disorders. She coordinates communications between the lab, study participants, providers, and collaborators and enrolls families into our research studies. Through the CCDD clinic, she provides genetic counseling for patients affected by cranial nerve disorders and their families. Brenda received her BA in Biology from Williams College and MS in Genetic Counseling from Brandeis University. Her doctoral research in the lab of Dr. Michael Fox focused on the neuroimmune mechanisms underlying cortical and hippocampal inhibitory circuit changes induced by long-term brain infection with the parasite,<span>\u00a0<\/span><em>Toxoplasma gondii<\/em>. She works closely with both the clinical neurogenetics and basic neuroscience research groups in the lab. She earned her PhD in Genetics from Fudan University in 2021, where she identified novel pathogenic genes in short stature patients through large-scale whole-exome sequencing and functional validation. In the Engle Lab, Hongdou is pursuing her goal of becoming a clinical geneticist by investigating the genetic causes of congenital cranial dysinnervation disorders (CCDDs). He conducted his doctoral research in the lab of Dr. Ghosh Roy on the regulation of microtubule cytoskeleton in <em>C. elegans<\/em> mechanosensory neuron. In the Engle Lab, his work focuses on studying the role of neuron specific \u03b2-tubulin, TUBB3, in cranial nerve development, and its interaction with the KIF21A motor protein, using cell biological and biochemical approaches. At Boston Children's Hospital, she is a member of the Departments of Neurology, Ophthalmology and Medicine (Genetics), a member of the F.M. Kirby Neurobiology Center and the Program in Genomics, and a senior investigator for The Manton Center for Orphan Disease Research. Dr. Engle received her B.A from Middlebury College and her M.D. from Johns Hopkins University School of Medicine. She trained in pediatrics at Johns Hopkins, in neuropathology at Massachusetts General Hospital, and in adult and child neurology in the Longwood Neurology Training Program and at Boston Children's Hospital. Following her residencies, she was a research fellow with Louis Kunkel, Ph.D., and later Alan Beggs, Ph.D., in the Division of Genetics at Children's prior to establishing her own research lab in 1997. Her work has defined the human congenital cranial dysinnervation disorders and has been recognized by high-profile publications and by receipt of multiple honors, including the E. Mead Johnson Award for Research in Pediatrics from the Society for Pediatric Research, the Sidney Carter Award in Child Neurology from the American Academy of Neurology, a Research Award for Vision from the Alcon Institute, and election to the National Academy of Medicine. She coordinates communications between the lab, study participants, referring providers and research collaborators in the US and abroad and enrolls families into our research studies. Through the CCDD clinic, she provides genetic counseling for pediatric and adult patients affected by cranial nerve disorders, as well as their families.<o:p><\/o:p>\n\nBrenda received her BA in Biology from Williams College and her MS in Genetic Counseling from Brandeis University. She is Board Certified by the American Board of Genetic Counseling and is a Licensed Genetic Counselor in the State of Massachusetts. She received her undergraduate degree in Molecular and Cell Biology from the University of Illinois at Urbana-Champaign in 2010.&nbsp; She then worked for two years in the laboratory of Dr. Pedro Jose at Children\u2019s National Medical Center (Washington, DC) and the University of Maryland School of Medicine (Baltimore, MD), where she researched mechanisms of blood pressure regulation. In 2017, she obtained her PhD in Human Genetics at Johns Hopkins University School of Medicine (Baltimore, MD) under the guidance of Dr. David Valle, where she identified novel candidate genes for unexplained Mendelian disorders through whole exome sequencing at the Baylor-Hopkins Center for Mendelian Genomics and performed mechanistic studies of a heritable bone dysplasia with retinopathy. He conducted his doctoral research in the lab of Don Conrad, PhD, studying the impact of rare copy-number variants on common infertility in humans and mice.<\/span>\n\nIn the Engle Lab, Dr. Arthur Lee\u2019s current work focuses on two major areas:\n<ol>\n \t<li>Identifying pathogenic structural variants (e.g., deletions, duplications, inversions, etc.) from patient whole genome sequencing data.<\/li>\n \t<li>Generating and analyzing single cell functional genomics data (e.g., scATAC-seq) for rare cell subtypes that are sparsely represented in public databases.<\/li>\n<\/ol>\nUltimately, he hopes to integrate these layers of next-generation sequencing data as a powerful lever into rare, non-coding disease biology. When not pursuing a cappella fame, he studied protein-trafficking in macrophage phagocytosis using FRET (fluorescence resonance energy transfer) imaging and live-cell microscopy. He went on to complete his medical training at the Warren Alpert Medical School of Brown University and completed a research fellowship at the&nbsp;National Institute on Deafness and Other Communication Disorders&nbsp;investigating novel targeted chemotherapeutics for head and neck squamous cell carcinoma.<\/p>\nDr. Mohan completed his general surgery internship at Brigham and Women\u2019s Hospital and is currently pursuing the seven-year research track in the Harvard Otolaryngology Residency program. His research is focused on Schwann cell plasticity and the peripheral nerve injury response with single-cell RNA sequencing of rodent and human neural tissues.&nbsp;<span>This work is conducted in collaboration with Drs. He received his BS in Chemistry at the University of Virginia, where he studied inner ear hair cell regeneration with Jeffrey Corwin, PhD. Matt earned his MD\/PhD in Developmental Biology in the lab of Huda Zoghbi, MD at Baylor College of Medicine in Houston, TX. There he mapped the fate of <em>Atoh1<\/em>-dependent brainstem neurons contributing to shared neural networks vital for balance, proprioception, hearing, and arousal.\n\nIn the Engle lab, Matt is using a combination of single cell and in situ transcriptomics, as well as whole organism tissue clearing and imaging to identify and visualize motor neuron subpopulations and their differential connectivity in health and disease.
